Título: EVALUATION OF MICRORNAS ASSOCIATED WITH THE AGGRESSIVE POTENTIAL OF SALIVARY GLAND MUCOEPIDERMOID CARCINOMA

Resumo: Objective: The aim of this study is to validate the use of microRNA expression for the discrimination of the metastatic potential of mucoepidermoid carcinoma (MEC) samples. Study design: Using Real Time RT-PCR (qPCR) we have analyzed the expression of 384 miRNAs and controls in 4 non-metastatic MECs, 3 MECs with lymph node metastasis, 3 MECs with distant metastasis, and 2 non-neoplastic human salivary gland samples. Comparative analysis identified 34 differentially expressed miRNAs potentially associated with metastatic behavior. For validation, 12 miRNAs were selected and evaluated in two independent MEC sample sets (10 and 13 samples) by qPCR. Results: Among the 12 selected miRNAs, miR-27a-3p, miR-133a-3p, and miR-134-5p were confirmed as differentially expressed. miR-27a-3p showed higher expression in non-metastatic tumors compared to metastatic groups. miR-133a-3p expression was higher in non-neoplastic samples than in cases with lymph node metastasis. In contrast, miR-134-5p expression was elevated in distant metastasis compared to lymph node metastasis. Conclusion: The determination of a miRNA expression profile could provide a better understanding of the molecular basis of MEC and contribute to the determination of diagnosis/prognosis of patients. Additionally, these findings may support the development of targeted therapeutic strategies. Supported by FAPESP (22/14253-7) and CNPq (303753/2025-0).
