Título: INSIGHTS INTO THE ANTITUMOR EFFECT OF GALLIC ACID IN A SARCOMA 180 MICE MODEL: NF-ΚB SUPPRESSION AND INTRINSIC APOPTOSIS

Resumo: Objective: This study investigated the antitumor effects and underlying mechanisms of gallic acid (GA) in a murine Sarcoma 180 (S180) model. Study design: Fifty male Swiss mice bearing dermal S180 tumors received oral GA (10, 25, or 50 mg/kg/day), 5-fluorouracil (25 mg/kg, i.p.), or vehicle for seven days. On day eight, animals were euthanized and tumors were collected for evaluation of tumor weight, growth inhibition, histopathological features, and immunohistochemical expression of proliferation (Ki-67), apoptosis (caspase-3, caspase-8, Bax, and Bcl-2) and ferroptosis (NRF2) markers, and the transcription factor NF-κB. Results: Treatment with GA at 50 mg/kg significantly reduced tumor weight (4.3 ± 0.3 vs. 11.3 ± 0.9 g/100 g; p<0.001), Ki-67 index (9.5 ± 3.79% vs. 14.2 ± 2.6%; p<0.001), and NF-κB expression (16.8 ± 7.7% vs. 38.4 ± 9.2%; p<0.001), compared with vehicle. In contrast, GA increased caspase-3 (9.0; 6.0-9.0 vs. 3.5; 2.0-4.0; p<0.001) and Bax (8.0; 6.0-9.0 vs. 3.0; 2.0-6.0; p<0.001) expression. No significant differences were observed in Bcl-2, caspase-8, or NRF2 expression (p>0.05). Conclusion: GA exerted antitumor activity in the S180 model, associated with inhibition of NF-κB signaling and activation of the intrinsic apoptotic pathway, supporting its potential as a candidate for adjuvant cancer therapy.
