Título: RPS6 IMMUNOEXPRESSION IN PALATINE MUCOSA OF RATS SUBJECTED TO 4NQO-INDUCED CARCINOGENESIS AND INDIRECT PBM

Resumo: Objective: to evaluate the RPS6 immunoexpression in palatine mucosa of rats previously exposed to 4NQO and subjected to distant photobiomodulation (PBM).
Study design: 33 samples were analyzed. The animals received systemic 4NQO during 20 weeks and were divided into three groups according to the laser protocol: control (n=11), PBM 3s (n=11) and PBM 10s (n=11). HE analysis showed SCC in all cases. Samples were submitted for immunohistochemical staining with RPS6 that was evaluated considering the percentage of positive cells in areas of normal epithelium, dysplasia and tissue invasion inside each case and among the groups. The proportion of positive cells was scored as 0 (<25%), 1 (25-50%), 2 (51-75%), or 3 (76-100%). Statistical analysis was conducted on Jamovi software.
Results: Differences in immunostaining were observed among the distinct epithelial alterations, with higher expression in normal epithelial mucosa and a progressive decrease in dysplasia and carcinoma (p<0.001). No significant differences were observed between groups with and without PBM exposure in normal mucosa (p=0.061), dysplasia (p=0.903), or carcinoma (p=0.524).
Conclusion: Loss of RPS6 immunostaining is associated with the acquisition of a malignant phenotype in oral carcinogenesis induced by 4NQO, regardless of PBM use.
