Título: INTEGRATED PROFILING OF AMELOBLASTOMA CELL LINES

Resumo: Objective: Ameloblastoma is a benign yet locally aggressive odontogenic tumour with high recurrence rates and frequent BRAFV600E mutations. Translational progress has been limited by the scarcity of well-characterised tumour cell models. This study aimed to define the phenotypic, molecular, and functional landscape of three ameloblastoma cell lines derived from the same tumour.
Study design: HAM1, HAM2, and HAM3 cell lines were analysed using whole-exome sequencing, RNA sequencing, and gene fusion detection. Migration and invasion assays were performed in all lines, while three-dimensional spheroid growth was assessed in HAM2 and HAM3. Candidate pathways were identified by gene set enrichment analysis and validated using qPCR and Western blotting.
Results: All cell lines harboured the BRAFV600E mutation, and no pathogenic gene fusions were detected. Transcriptomic profiling revealed heterogeneity and enrichment of epithelial-mesenchymal transition (EMT) and KRAS-related gene sets. HAM3 demonstrated greater migratory, invasive, and spheroid-forming capacity than HAM2. EMT-associated markers CDH11 and TGM2 were overexpressed at both gene and protein levels.
Conclusions: These findings reveal intra-tumour heterogeneity and identify EMT-associated pathways as potential drivers of ameloblastoma behaviour, supporting their relevance as therapeutic targets and reinforcing the value of ameloblastoma-derived cell models for precision oncology research.
