Título: EXTRACELLULAR MATRIX IN MALIGNANT SALIVARY GLAND TUMORS: A STUDY USING SPECIAL STAINS AND SECOND HARMONIC GENERATION

Resumo: Objective: To identify and compare extracellular matrix (ECM) components in malignant salivary gland tumors using special staining (SS) and second harmonic generation (SHG).Study Design: Twenty malignant salivary gland tumors were analyzed, including adenoid cystic carcinoma (ACC), myoepithelial carcinoma (MC), epithelial-myoepithelial carcinoma (EMC), and salivary duct carcinoma (SDC). SS was performed for elastin, collagen fibers (CF), reticular fibers (RF), proteoglycans (PG), and glycosaminoglycans (GAG). SHG assessed CF parameters: area fraction (AF), gray level (GL), contrast, and entropy. Data were analyzed using ANOVA and Mann-Whitney tests (p < 0.0005). Results: SS revealed significantly higher CF in ACC, SDC, and EMC compared to MC. RF levels were higher in MC and EMC than in SDC. SDC showed increased PG and GAG compared to MC. Elastin showed no significant differences. In SHG, AF was higher in ACC, MC, and EMC versus SDC. GL was elevated in MC, ACC, and EMC relative to SDC. Contrast was highest in MC and EMC, with ACC also surpassing SDC. Entropy was significantly higher in ACC, MC, and EMC compared to SDC. Conclusion: ECM composition differs among malignant salivary gland tumors, highlighting its potential role in tumor behavior and its relevance as a target for future therapeutic strategies.
