Título: Identification of Novel Therapeutic Inhibitors for Head and Neck Cancer in Young Patients via High-Throughput Screening

Resumo: Objective: This study aimed to perform a high-throughput drug screening using a comprehensive library of FDA-approved drugs to identify potent and selective candidates for the treatment of head and neck squamous cell carcinoma (HNSCC) in young patients.Study design: A total of 2,320 FDA-approved drugs were screened against the ATCC-SCC9 cell line, derived from a 25-year-old male patient with tongue squamous cell carcinoma. Cells were robotically seeded at a density of 2,000 cells per well in 384-well plates and incubated for 24 hours before drug exposure. Each compound was administered at a final concentration of 10 μM using an automated dispensing system. After an additional 24-hour incubation, cell viability was assessed using propidium iodide staining.Results: Thirty-six hit compounds were identified as significantly reducing SCC9 cell viability. These included a diverse set of agents such as multi-targeted inhibitors of histone deacetylases, EGFR, NF-κB, and HER2; natural products; antimalarial, antiarrhythmic, antiviral, and antibiotic agents; a drug for psoriasis; and anthracycline-based antineoplastic agents.Conclusion: This high-throughput screening identified a range of FDA-approved drugs with potential efficacy against HNSCC. These findings offer a promising starting point for the repurposing and further investigation of existing therapeutics to manage HNSCC, especially in younger patient populations.
