Título: EVALUATION OF BRAF p.Val600Glu MUTATION IN ADENOMATOID ODONTOGENIC TUMOR

Resumo: Objective: KRAS codon 12 mutations occur in 70% of adenomatoid odontogenic tumors (AOT). Recently, BRAF p.Val600Glu mutation has been reported by a single study in two AOT cases, one of which also harbored KRAS p.Gly12Val. Given that BRAF p.Val600Glu had not been previously detected in AOT and that BRAF and KRAS mutations are considered mutually exclusive, we aimed to investigate the presence of BRAF p.Val600Glu in both KRAS-mutated and wild-type AOT cases, and to explore the potential co-occurrence of BRAF and KRAS mutations. Study Design: gDNA was isolated from 12 formalin-fixed, paraffin-embedded (FFPE) AOT samples with known KRAS mutational status. 7/12 cases were KRAS mutation-positive and 5/12 were KRAS wild-type. BRAF p.Val600Glu was assessed using allele-specific qPCR and/or Sanger sequencing. Results: None of the samples tested positive for BRAF p.Val600Glu; therefore, no co-mutation was observed. Conclusion: BRAF p.Val600Glu mutation was not detected either in the KRAS wild-type or in the KRAS mutation-positive cases, suggesting that BRAF is not involved in the pathogenesis of the analyzed samples.
