Título: ADVERSE EFFECTS OF ELECTRONIC AND CONVENTIONAL CIGARETTES ON THE MYCOBIOME AND ORAL VIROME

Resumo: Oral microbiome dysbiosis has been associated with various oral and systemic pathologies, often driven by external factors such as smoking. With the increasing prevalence of e-cigarette use, concerns have emerged regarding their potential impact on oral health. This study aimed to evaluate the effects of conventional cigarette use, e-cigarette use, and dual use on the composition of the oral mycobiome and virome by reanalyzing publicly available metagenomic data. Sequencing data from BioProject PRJNA544061 were preprocessed and analyzed using Bowtie2, Kraken2, and Bracken, with interactive visualizations generated in Pavian. Fungal and viral diversity and abundance were compared across user groups. The greatest fungal diversity was observed in individuals using both conventional and electronic cigarettes, with a predominance of Candida albicans and Malassezia restricta, related to increased intensity of inflammation of the oral mucosa. In the virome, notable increases in Streptococcus phage SM1, S_Latrobevirus_FN1, Dybvigvirus_AV1, and S_Aeromonas_phage_LAh10 associated to increased virulence of Streptococcus Mutans, Fusobacterium Nucleatum and Actinomyces, were particularly evident among former smokers who transitioned to e-cigarettes. Overall, the use of tobacco products-whether conventional, electronic, or both-significantly alters the oral microbial environment, promoting dysbiosis and potentially contributing to adverse clinical outcomes.
