Título: IMPACTS OF E-CIGARETTE USE ON TP53, INFLAMMATORY, AND KERATINIZATION PATHWAYS IN THE ORAL EPITHELIUM

Resumo: Objective: This study aimed to evaluate gene expression patterns related to inflammatory response, DNA repair, and epithelial keratinization in the oral mucosa of electronic cigarette (e-cigarette) users. Design: Forty-two volunteers were recruited and equally divided into two groups: a control group, consisting of individuals who had never used conventional or electronic cigarettes, and an e-cig group, comprising exclusive users of vaporization devices. Sociodemographic data, including on sex, age, and frequency of electronic cigarette use, were recorded. Oral mucosa samples were collected from the lateral border of the tongue using exfoliative cytology. Total RNA was extracted, and qPCR was performed to assess the expression of performed to assess the expression of IL-1B, CXCL8, TNF, KRT1, KRT13, KRT19, TP16, TP21, and TP53. Results: Most genes were downregulated in the e-cig group compared to controls, except for TP53, which was significantly upregulated. Notably, gene expression correlations were observed between KRT13-KRT19 and TP21-TNF. Male predominance was identified among frequent e-cigarette users. Conclusions: These results highlight the vaporization effects on the oral mucosa and underscore the importance of conducting more extensive cohort studies to further investigate the potential long-term effects of electronic cigarettes and their derivatives.
