Título: ANTITUMOR EFFECT OF ARRABIDAEA CHICA EXTRACT IN SARCOMA 180: A NATURAL AGENT TARGETING MITOCHONDRIAL APOPTOSIS

Resumo: Objective: This study evaluated the antitumor potential of the hydroethanolic extract of Arrabidaea chica Verlot leaves (HEAC), focusing on the potential intrinsec apoptotic pathway activation. Sudy design: HEAC was obtained via ultrasound-assisted pressurized liquid extraction and characterized by HPLC. Then, swiss mice bearing sarcoma 180 tumors were treated for seven days with saline, 5-fluorouracil (5FU, 25 mg/kg), HEAC (100, 200, or 300 mg/kg), or combinations of HEAC with 5FU. Results: Rutin, quercetin, luteolin and Kaempferol were the major compounds found in HEAC. HEAC at 200 and 300 mg/kg significantly inhibited tumor growth (p<0.01), with no difference between these doses; thus, 200 mg/kg was selected for further analysis. Immunohistochemistry revealed that HEAC significantly increased TUNEL, Bax, and active caspase-3 expression (p<0.05), suggesting mitochondrial apoptotic pathway activation. No changes were observed in Ki67, NF-κB, or NRF2 expression, suggesting that HEACs antitumor action is not linked to proliferation arrest, NF-κB modulation, or ferroptosis. The HEAC/5FU combination did not enhance efficacy compared to single treatments. Conclusions: These results highlight HEAC (200 mg/kg) as a promising natural antitumor agent acting through apoptosis activation, supporting its potential for further development in cancer therapy. Additional studies are warranted to clarify its molecular mechanisms and therapeutic applicability.
