Título: ANALYSIS OF PYROPTOSIS AND NECROPTOSIS INDUCED BY DYNAMIC THERAPIES IN DYSPLASTIC ORAL KERATINOCYTES: AN IN VITRO STUDY

Resumo: Objective: To investigate pyroptosis and necroptosis induced by photodynamic (PDT) and sonodynamic (SDT) therapies on dysplastic oral keratinocytes (DOK) in vitro. Methods: DOK cells were incubated with 5-aminolevulinic acid (5-ALA) for 4h in the dark, followed by exposure to LED light (630±5nm, 150mW) for 1, 3, or 5min (3, 9, or 15J/cm²). SDT was performed using 1MHz ultrasound in pulsed mode (50% duty cycle) for the same durations. For combined sono photodynamic therapy (SPDT), both sources were applied simultaneously. Cell viability was assessed by MTS, membrane rupture by LDH release, and pyroptosis and necroptosis by immunofluorescence of GSDMD and MLKL. Results: PDT showed a dose-dependent cytotoxic effect. SDT reduced viability without a clear dose-response. SPDT caused significantly greater cell death at all durations. LDH release was highest after SPDT, and immunofluorescence revealed significantly increased GSDMD and MLKL expression in SPDT-treated DOK cells. Conclusion: SPDT mediated by 5-ALA is more effective than PDT or SDT alone in inducing pyroptosis and necroptosis in dysplastic oral keratinocytes. These findings support SPDT as a promising therapeutic approach for oral epithelial dysplasia.
