Título: Evaluation of Anti-Angiogenic Agents and NFκB Sensitization in Adenoid Cystic Carcinoma Cells

Resumo: Objective: Conventional therapies, such as surgery, chemotherapy, and radiotherapy, are not always effective for the treatment of Adenoid Cystic Carcinoma (ACC). Given these limitations, the present study evaluated the antiangiogenics, Axitinib, Everolimus, and Lenvatinib, alone and in combination with the NFκB inhibitor Emetine, on ACC cell viability.
Study Design: ACC cells were cultured and treated with antiangiogenics. Cell death was measured by Propidium Iodide and Hoechst staining. Immunofluorescence assays were used to detect NFκB in vitro. NFκB activation was confirmed in vivo in PDX tumors with immunofluorescence and quantified with ImageJ. Cell death was re-evaluated following exposure to Emetine.
Results: Antiangiogenics reduced cell viability in a dose-dependent manner, with IC50 values of 2.6 µM (Axitinib), 29.02 µM (Everolimus), and 23.76 µM (Lenvatinib). However, a longer treatment period with these drugs enhanced the expression of NFκB, which was also confirmed through fluorescence analysis of PDX slides. Emetine inhibited NFκB activation and, when combined with antiangiogenic agents, synergistically enhanced cytotoxicity, leading to significantly increased cell death.
Conclusion: These findings emphasize the possibility of combining NFκB inhibitors and antiangiogenic agents to address adaptive resistance in ACC, offering a novel and promising approach for improving therapeutic efficacy against this challenging cancer.
