Título: ABNORMAL p53 IMMUNOHISTOCHEMICAL PATTERNS IN ORAL POTENTIALLY MALIGNANT DISORDERS AND ORAL CANCER

Resumo: Objective
To evaluate a large case series of oral leukoplakia (OL), proliferative verrucous leukoplakia (PVL), actinic cheilitis (AC), lower lip squamous cell carcinoma (LLSCC), conventional oral squamous cell carcinoma (OSCC), oral verrucous carcinoma (OVC), and inflammatory fibrous hyperplasia (IFH), aiming for a comparative analysis and determining the frequency of wild-type and abnormal p53 immunohistochemistry (IHC) patterns in these lesions.

Study Design
The samples were divided into groups: OL (n=57), PVL (n=54), AC (n=32), LLSCC (n=25), OSCC (n=34; of them, 10 PVL-derived SCCs), OVC (n=31), and inflammatory fibrous hyperplasia (IFH) (n=17).

Results
When comparing the groups, 17% (25/143) of OPMDs and 37% (22/59) of LLSCC/OSCCs presented an abnormal p53 pattern (p<0.05). Within the OSCC group, 20% of PVL-derived OSCC and 70% of non-PVL-derived OSCC presented abnormal p53 patterns (p<0.007). Notably, only 3% (1/31) of OVCs exhibited abnormal p53 patterns, different from LLSCC (12%) (p=0.205) and OSCC (56%) (p<0.0001). No association was found between the epithelial dysplasia (oral potentially malignant disorders [OPMDs])/histological grading (SCCs) systems and abnormal p53 patterns (p=0.082).

Conclusion
Our results suggest abnormal p53 pattern-OPMD/SCC subgroups, independent of differentiation degree, which appear to have a therapeutic and prognostic impact.
