Título: INVESTIGATION OF GINGIVAL TISSUE AS A LATENT HIV RESERVOIR: PRELIMINARY DATA FROM A NON-RANDOMIZED CLINICAL TRIAL

Resumo: Objective: To analyze whether the gingival tissue (GT) is a reservoir for HIV-1 and the potential influence of periodontitis.

Study Design: GT biopsies and peripheral blood mononuclear cells (PBMC) were collected from HIV-1-infected patients treated successfully by antiretroviral therapy (ART) without (n=26) and with (n=26) periodontitis before and after non-surgical periodontal treatment (NSPT), along with periodontal and immunological parameters. HIV DNA load in the samples was evaluated by Real-Time PCR. This study was approved by a Research Ethics Committee (CAAE: 81355517.1.0000.5419), patients provided free informed consent.

Results: HIV DNA was detected in the GT of 9 out of 52 patients (17.3%), without association with their periodontal status. Positivity for HIV DNA in GT was associated with the presence of high proviral DNA in PBMC (p <0.05 Mann-Whitney U test). Significant improvement in all periodontal parameters was observed after NSPT (p<0.001 Students t test).

Conclusion: Proviral DNA was detected in the GT of some subjects living with HIV and treated by ART, independent of periodontal status. Additional studies are ongoing to explore the potential role of GT as a reservoir for HIV.
