Título: SCD-1 AND SCLEROSTIN IMMUNOEXPRESSION IN ORAL SQUAMOUS CELL CARCINOMA

Resumo: This study aimed to evaluate the immunoexpression of Stearoyl-CoA Desaturase-1 (SCD-1)
and Sclerostin in Oral Squamous Cell Carcinoma (OSCC), and to compare cases based on their
proximity to the bone. Thirty-nine cases were included and categorized based on their proximity
to the bone: NT (next to the tumor in the gingiva, alveolar ridge, palate, and T3/T4 floor of mouth
and tongue) and away from the lesion (AT, T1/T2 of the tongue and floor of the mouth, and
oropharynx). Immunoexpression was assessed using two semi-automated methods: QuPath®
for detecting total positive cells and staining intensity, and Image J/Fiji® for color deconvolution
and pixel analysis. Statistical analysis was performed using Mann-Whitney and Kruskal-Wallis
tests. SCD-1 exhibited weak immunoexpression in 89.7% and intense expression in 2.6% of
cases, while sclerostin showed weak expression in 57% and intense expression in 6% of neoplastic cells.
Both proteins demonstrated lower pixel intensity for the NT group. Notably, SCD-1 was more
strongly expressed in deeper tumor cells (p<0.01), while sclerostin was more intense in
superficial tumor cells (p<0.01). The findings suggest that SCD-1 and sclerostin are expressed
in OSCC, particularly in tumors near bone structures, and may play a role in tumor progression
and microenvironment.
