Título: IMMUNOHISTOCHEMICAL ANALYSIS OF LIPOGENESIS IN A LARGE CASE SERIES OF HEAD AND NECK SQUAMOUS CELL CARCINOMA

Resumo: Objective
Cancer cells frequently reprogram metabolic pathways for survival, proliferation, and metastasis. Although the glycolysis pathway is well studied, the role of lipid metabolic reprogramming in supporting cancer growth and proliferation, especially metastasis, is less understood.
Study Design
498 cases of head and neck squamous cell carcinoma (HNSCC), including oral (OSCC; n=237, of them, 31 were verrucous carcinomas [VCs]), oropharyngeal (OPSCC, n=115), laryngeal (LarSCC, n=88), Lip (LipSCC, n=44) and sinusal (SSCC, n=14) squamous cell carcinomas, were evaluated through CD36, perilipin, adipophilin, GCDFP-15 and mammaglobin immunomarkers, in correlation with clinicopathological parameters, including HPV infection status. The results were subjected to statistical analysis.
Results
When comparing HNSCCs in relation to HPV status, only CD36 and adipophilin showed significant association. Relevantly, only CD36 showed significant association, when comparing lipogenic markers with lymph node metastasis. Only adipophilin showed significant association with chemotherapy and radiotherapy. Without significant association with recurrence, only CD36 showed a trend towards significance (p=0.075). Moreover, only VCs showed absent/low expression of lipogenic markers.

Conclusion

Our results show differential expression of CD36 and adipophilin in HNSCCs, with therapeutic and prognostic impact, especially considering metastasis and (neo)adjuvant treatment. The excellent prognosis of VC can be explained by the absent/low expression of lipogenic markers.
