Resumo: This study explores the role of cellular senescence in head and neck squamous cell carcinoma (HNSCC) and evaluates the therapeutic potential of senescence-targeted interventions. Cellular senescence is a tumor-suppressive mechanism that prevents the proliferation of damaged cells but also contributes to chronic inflammation and tumor progression through the senescence-associated secretory phenotype (SASP). A narrative review was conducted to analyze recent findings on senescence regulation in HNSCC, with a focus on induced senescence therapy. Evidence suggests that inducing senescence in tumor cells may enhance treatment efficacy; however, the pro-inflammatory effects of SASP present challenges, potentially promoting an immunosuppressive microenvironment and tumor recurrence. Emerging therapeutic strategies, including senolytics and senostatics, aim to selectively eliminate or modulate senescent cells to harness their benefits while mitigating adverse effects. These approaches show promise in improving treatment outcomes by balancing senescence induction with controlled immune responses. While induced senescence therapy represents a novel strategy for HNSCC, its clinical application requires precise modulation to optimize therapeutic benefits and minimize risks. Therefore, further research is needed to optimize these approaches and incorporate them into precision oncology, potentially advancing HNSCC treatment by refining our understanding of senescence dynamics in cancer therapy.
