Título: CD8+ T CELLS AND GALR2 EXPRESSION IN PBMC OF ORAL SQUAMOUS CELL CARCINOMA (OSCC) PATIENTS

Resumo: Objective: GALR2 is considered an oncogene in OSCC and CD8+ T cells are associated with anti-tumoral responses and better prognosis. The purpose was to investigate the prevalence of CD8+ T cells and GALR2 expression in peripheral blood mononuclear cells (PBMCs) of OSCC patients.
Study Design: Venous whole blood from 16 OSCC cases (n=8 T1/T2 and n=8 T3/T4) was collected at the moment of surgical resection of tumors and PBMCs were isolated. Flow cytometry was performed to identify CD3+CD8+ T cells. Prevalence of CD8+ T cells was correlated with TNM clinical staging. RT-qPCR was performed using RNA extracted from aliquots of the same PBMC samples of a subgroup of these patients (n= 6) to determine expression of GALR2.
Results: There was non-statistically significant trend of increased number of CD8+ T cells in OSCC patients with larger tumors (T3 or T4). GALR2 expression was significantly greater (p=0.005) in samples with higher number of CD8+ T cells. There was no association between tumor size and GALR2 expression in PBMC.
Conclusion: Larger OSCC tumors tend to be associated with increased number of CD8+ T cells in the peripheral blood. GALR2 expression in PBMC was greater in cases with higher numbers of CD8+ T cells.
